The man asking the question is represented by Venus, 1st house ruler, in a very weakened condition: in exile in Aries and combust.

His wife is represented by Mars, peregrine, so not in a power position either, although stronger than Venus.

The alleged affair of the wife would be represented by the Sun, ruler of the 5th house (love) from the 7th (wife).

There are several indications in this chart which ultimately lead to the 'yes' answer:

the Moon sextile Sun's antiscia (05Vir13) - this an aspect between the main significators
the Moon afflicts by square the Ascendant-Descendant axis - that is the husband-wife relationship
no direct aspect, collection or translation of light between the 1st and 7th house rulers, Venus and Mars, that would indicate a strong marital relationship
Venus (husband) receives Mars by term (he loves/cares about her), but Venus is not received back in Mars' term (but in Saturn's; keyword: solitude)
separating aspect between the 1st and 7th house Almutens, Saturn and the Sun, indicating a weakening in the relationship (Almuten = the planet which has most essential dignities at a house cusp's degree, sometimes more than the common house ruler)
the Sun is both the ruler of the turned 5th house from the 7th (wife's affair) and Almuten of the 7th (wife)

These indications allow the conclusion that the wife is deceiving him.
